Vaccine treatment for Type 1 Diabetes shows promise

After a several years of study and trials, the Type 1 Diabetes Vaccine has reached a new milestone thanks to Doctor Doug Melton. Dr. Melton has helped to create a new innovative way to treat and even vaccinate against Type 1 Diabetes using stem-cell research. He started the work over 10 years ago, when stem-cell research was raising hopes and controversy. In 2014 he co-founded Semma Therapeutics—the name is derived from Sam and Emma—to develop the technology, and this summer it was acquired by Vertex Pharmaceuticals for $950 million. The company has created a small, implantable device that holds millions of replacement beta cells, letting glucose and insulin through but keeping immune cells out. “If it works in people as well as it does in animals, it’s possible that people will not be diabetic,” Melton says. “They will eat and drink and play like those of us who are not.”

Human trials of new Alzheimer’s treatment begin

The New York Medical University has reported today that it is now starting human trials of a new Alzheimer’s treatment working in co-junction with Biogen based in Cambridge, Massachusetts. Biogen hopes that the trials will be successful and allow the company to move forward with plans to to submit a Biologics License Application to the FDA for its anti-amyloid antibody, aducanumab. If approved, it will be the first new drug available to patients with Alzheimer’s disease in 16 years.

Administered intravenously once a month, aducanumab clears some of the plaque materials that may cause cell death and tissue loss in the brain caused by Alzheimer's disease, according to Barry Reisberg, MD, one of the clinical investigators who developed the Alzheimer’s drug memantine, which was approved back in 2003. Although it is not curative, aducanumab could be a valuable addition to the clinician’s armamentarium, according to Sharon A. Brangman, MD, chair of the geriatrics department and director of the Center for Excellence for Alzheimer’s Disease at Upstate Medical University in New York.

“Aducanumab is exciting in terms of adding more information to our knowledge of Alzheimer’s disease and maybe offering potential treatment, but it is not a cure,” she said.

Type 1 Diabetes vaccine enters new phase

Doctor Doug Melton, the man leading the charge against curing or at least preventing Type 1 Diabetes has reported that the testing done so far has shown great promise. His report states researchers tested the safety and feasibility data of a first-in-man-prospective open-labeled, placebo-controlled, dose-escalation phase 1 trial in nine people with long-standing type 1 diabetes (for at least 12 years)



The researchers aimed at investigating the use of tolerogenic dendritic cells (TolDCs) and proinsulin peptide (the precursor to insulin) to intervene in the pathogenesis of type 1 diabetes. Tolerogenic dendritic cells are a type of immune responding cell that may be able to manage the beginning of underlying autoimmunity at the time-at-onset and onwards.

Participants were administered two intradermal vaccinations series (5, 10, or 20, depending on dose cohort) of TolDCs pulsed with proinsulin peptide, one month apart. During the six months of intensive monitoring, the researchers found that all participants diabetes control remained stable and that the intervention appeared to be safe and well-tolerated, with few adverse reactions.

Although they did not “cure” the participants of diabetes, there was no acceleration of beta cell loss during the intervention. They concluded that their study warrants “Subsequent clinical testing in patients with a shorter diagnosis of type 1 diabetes (less than 12 years) and with preserved C-peptide production, to assess whether this novel immune intervention strategy is able to delay or halt progressive loss of beta cell function.” Another study to recently hit the news approaches a vaccine for type 1 diabetes in an alternative way. As mentioned, it has been discovered that certain viruses are linked to the development of type 1 diabetes, one speculated culprit, Coxsackievirus B (CVB) enteroviruses. There are six different strains of this virus, all of which are linked to different diseases.

Researchers formulated a vaccine for all six strains of CVB and found that it had positive results on both mice and monkeys. When given to monkeys, the vaccine stimulated antibodies to CVB, which implied that if it could prevent the development of CVB then it could prevent diabetes.6

While this research is certainly promising, there are a few drawbacks to the study. First off, the study was done in animals which has its own limitations.

In addition, it is difficult to determine if children who are predisposed to type 1 diabetes will develop the disease after exposure to a different type of virus or infection. More research will be needed to determine if these vaccines are safe and applicable in children.

The good news is that if they are, there is a chance they can prevent the cases of diabetes in children that are potentially caused by CVB.

The government of the USGA has no stake in the situation between Porte cale and Spain over this matter. However, since we have been asked to give our opinion on it, the USGA believes that any actions on the question should be delayed until there is a stable, legally accepted and functioning government in Spain to conduct negotiations with.

The USGA believes that Porte cale should take no action until that is a reality.

Tuscany is now under the control of Papal, Kommerian and Pisan forces. The task now is the mopping up of any remaining Communist forces left, the reestablishing of basic services to the population, and the resting, resupply and bringing up to strength of the Papal forces.

In the matter of the hundreds of prisoners taken, many of them have stated that they were not really Communists, but had been forced into the PRI militia against their will. While many of these claims are no doubt exaggerated, Hos Holiness Pope Celestine VI has decreed that all those, excepting the leaders of the Communists, who are willing to take an oath of allegiance to himself, the Holy Mother Church, and the Papal States are to be released and allowed to return to their homes. Any who wish to join the Papal Forces will be allowed to. The leaders and the hard core Communists will be held in custody until such time as it is determined what to do with them.

In addition to re establishing basic services, and keeping law and order, the Papal Forces are currently involved in counting and cataloging the large quantities of military equipment captured from the Communists. It is expected that whatever is in condition to be put back into service will have that done, to be used again against their former owners.

The talks with the Republic of Pisa and Kommeria about the status of Pisa, and now of Tuscany, are still ongoing, and with the arrest of the Commander in Chief of the Kommerian forces for war crimes, the future of the Kommerian presence in the conflict is now in question.

With the arrival of winter, it is expected that no significant military activity will be undertaken. but that hasn't been definitely stated to be the case. Defense Minister Enzo Conti will be flying to The Eastern Latins in the next few days, although the purpose of his visit hasn't been released. Informed sources say that it may have something to do with the increased Byzantine military activity in the Western Mediterranean in recent weeks.

The democratic process has triumphed once again, and by an overwhelming majority, the moratorium on the 30 day non posting rule has been extended for 30 days, and will expire on 1 February 2021.

The official Tass News Agency is reporting that a revolt has broken out among the Moldovan troops of the 47th Motor Rifle Division in southern Moldova. The Moldovan capital of Chisinau has been wracked for several days by massive protests against the Government's move of troops to intimidate the breakaway Romanian province of Greater Austrias. It was feared that Moldovan soldiers of the Imperial Army would support the protestors. This was the reason given as to why the 146th Motor Rifle Regiment of the 47th, which is over 80% Moldovan in composition, had been locked down on its base. The fear that this very thing might happen. When several soldiers were arrested for being drunk and violating the lockdown, that was the spark that ignited the flame. Whole units revolted, armories and storage areas were taken over, and firefights broke out between the rebels and loyal troops. Other reports indicate that many rebel soldiers have taken to the woods, bringing their arms with them.

The 7th Army, which controls the forces in Moldova, has sent regiments of the 43rd Motor Rifle Division, along with the 37th Air Assault Brigade, 2 mostly Slavic units, to the base to quell the revolt, and severe fighting is anticipated before the revolt is crushed.

Pravda is reporting that the revolt in Moldova is spreading. The capital city of Chisinau is quiet for now, with soldiers of the 37th Air Assault Brigade joining the National Guard and police in enforcing a dusk to dawn curfew and a ban against gathering of more than 10 people, but there are reports of severe fighting in the south, with guerrilla attacks against army units near the city of Cimislia, and more Moldavan units of the Army joining the rebels. The Ministry of Defense has ordered more troops to be sent, among them the 108th and 247th Airborne Regiments of the 7th Imperial Banner Mountain Airborne Division.

It appears that the revolt is more widespread than previously thought, and the Government seems to only now be taking it seriously. A spokesman for the Foreign Ministry blamed the rebel Romanian government for provoking the revolt, and accused them of supplying arms to the rebels. A spokesperson for the Romanian rebels called this "Utter nonsense", saying "we have only enough arms for ourselves. We sympathize with our brothers and sisters in Moldova in their struggle for liberty, but we cannot give them anything more than moral support."
